The three directions out of CMB
The airport sits north of Colombo, which means three sensible first moves: north-east to the Cultural Triangle, east into the hills towards Kandy, or south along the expressway to the beaches.
All three avoid backtracking if you plan the rest of the trip as a loop, so the choice is mostly about your landing time and how much driving you want on day one.
North-east to Sigiriya and Dambulla
About 150 km and three and a half hours, and the only direction that skips Colombo traffic entirely. If you land in the morning you can be at your hotel by mid-afternoon and climb Sigiriya or Pidurangala at sunrise the next day.
This works particularly well for a clockwise loop: Cultural Triangle first, then Kandy, tea country, the south coast and back up the expressway to the airport.
East to Kandy
Kandy is 130 km inland and about three and a half hours because the last stretch climbs and winds. Many travellers turn it into a half-day by stopping at Pinnawala and a Matale spice garden.
It suits arrivals before early afternoon. Arriving in Kandy after dark means missing the lake and the hill views that make the drive worthwhile.
South to Bentota and Galle
The Southern Expressway puts Bentota two hours away and Galle two and a half. It is the fastest long transfer in the country and the easiest on jet lag, since most of it is smooth dual carriageway.
Pick this if the trip is beach-led, or if you are landing late and want a short second day.
If you land at night
For anything after about 8pm, stay near the airport. Negombo is twenty minutes from arrivals and gives you a beach for the morning; Katunayake and Seeduwa are closer still if you only need sleep.
Starting the long drive fresh the next morning is almost always more comfortable than arriving at a hill hotel at 2am, and it costs you no holiday time in practice.

Can I go straight from the airport to Ella?
Yes, but it is a five-and-a-half hour climb. Unless you land early, an overnight nearer the coast first is usually more comfortable.
Is it better to start north or south?
Either works as a loop. North-east to Sigiriya avoids Colombo traffic on day one; south on the expressway is the least tiring drive.
